Concerning the stability issue of gp 125, I am very interested to know if there is any kind of appropriate coating procedure for gp 125 to achieve better results and good stability. I would very much appreciate your guidance in this regard.
Resolution:
The best advice that I can give you is to be careful with the storage of the frozen antigen - minimizing freeze/thaw cycles as well as the time it remains thawed prior to dilution and coating.
Greater than 70% of the antigen will adsorb to the plastic or nitrocellulose within the first 60 minutes of absorption time. Once it absorbs to a solid surface, stability is greatly enhanced. Further enhancements of the stability may be obtained by using our DryCoat immunoassay stabilizer.
We usually use PBS as the coating diluent and NUNC polysorp plates.
