Question:
- How much does it cost if we don't need additional eggs or extended maintenance?
- How much of antigen do we need to provide for immunization?
- Is any conjugation or modification necessary when using peptides as antigens?
- If so, what do we need to prepare?
- In terms of purity, is a peptide with >70% HPLC purity good enough?
- Is it possible to receive some eggs sooner than the end of 12-week period
Resolution:
I have addressed each of your questions below:
- How much does it cost if we don't need additional eggs or extended maintenance? Total price for the animal work is $575 for 2 hens - 3 pre and 6 hyperimmune eggs from each
- How much of antigen do we need to provide for immunization? Approximately 1 milligram total - we need 5 vials of 150 - 200 µg each
- Is any conjugation or modification necessary when using peptides as antigens? Yes - peptides are too small to elicit an immune response. Conjugation is to KLH and is monitored by a quantitative Ellmans assay.
- If so, what do we need to prepare? A peptide with a terminal reduced cysteine. We can make this for you @ $23/residue if you like
- In terms of purity, is a peptide with >70% HPLC purity good enough? 70% is fine for immunizations
- Is it possible to receive some eggs sooner than the end of 12-week period? The 10 - 12 week period includes the time needed for purification. We can get you hyperimmune eggs at roughly week 7 - 8.
